Sanofi and Regeneron launched a major immuno-oncology collaboration in 2015, with Sanofi committing up to $2.17B including $640M in upfront payments. This expanded their long-standing antibody alliance that originated in 2003 and had produced Dupixent (dupilumab), Praluent (alirocumab), and other antibody therapeutics. The I-O collaboration was later restructured in 2019 when Sanofi paid $462M to exit.

Dupixent (dupilumab) was the first biologic for moderate-to-severe AD, inhibiting IL-4/IL-13. The Sanofi-Regeneron alliance produced one of pharma's most successful launches, expanding to asthma, CRSwNP, EoE, and prurigo nodularis.
Dupixent competes with JAK inhibitors upadacitinib (Rinvoq, AbbVie) and abrocitinib (Cibinqo, Pfizer), plus the IL-13-targeted biologic tralokinumab (Adbry, LEO Pharma) and lebrikizumab (Ebglyss, Eli Lilly). Topical ruxolitinib (Opzelura, Incyte) serves milder disease; nemolizumab (Nemluvio, Galderma) targets IL-31 pruritus signaling.
Sanofi committed up to $2.17B including $640M upfront for broad IO collaboration including cemiplimab.
Regeneron acquired exclusive global rights to cemiplimab from Sanofi for $900M upfront.
Libtayo (cemiplimab) approved for CSCC, NSCLC, cervical cancer. On track for $1B+ annual sales. Dupixent also massively successful.
