Sanofi acquired Amunix Pharmaceuticals for its Pro-XTEN masking technology platform, which enables conditionally activated biologics for cancer immunotherapy. The XPAT and XPAC platforms create masked T-cell engagers and pro-cytokines (IL-2, IL-15, IL-12) that activate preferentially in the tumor microenvironment, reducing systemic toxicity.
Amunix XTEN technology addresses a critical challenge in solid tumor immuno-oncology: systemic toxicity from T-cell engagers and cytokines. AMX-818 (HER2 masked TCE) is the lead program. XPAT platform produces masked bispecific T-cell engagers; XPAC produces masked pro-cytokines. Competitors include Cytomx (probody), Inhibrx, and Merus in conditional activation.
