Therapeutic Human Polyclonals (THP) was a platform company, not a single-indication therapeutics developer. Its core technology was the first transgenic rabbit system engineered to produce fully human antibody repertoires — both monoclonal and, distinctively, polyclonal antibody mixtures. Polyclonals are particularly attractive for infectious disease, transplant rejection, and toxin neutralization, where attacking multiple epitopes simultaneously may improve potency and resistance profile.
By 2007 the human-antibody platform space had three established leaders making fully human monoclonals: Medarex's HuMAb-Mouse and KM Mouse, Abgenix's XenoMouse (acquired by Amgen in 2006 for $2.2B), and Regeneron's VelocImmune. Phage-display competitors included Cambridge Antibody Technology (acquired by AstraZeneca in 2006) and Dyax. THP's differentiation was the rabbit host — historically prized for higher affinity and broader epitope coverage than mouse — combined with a polyclonal output not addressable by murine or phage platforms. Polyclonal therapeutics historically came from human plasma (IVIG, hyperimmune globulins) or animal sera (anti-thymocyte globulin, anti-venoms); a transgenic-rabbit-derived fully human polyclonal could in principle replace these with a more controlled, scalable, lower-immunogenicity product. For Roche, the $56.5M deal slotted into a broader 2007 buying spree (454 Life Sciences for $154.9M closed within days) aimed at platform breadth, with THP integrated into the Roche Protein Research Center in Penzberg, Germany. Strategic logic: cheap optionality on a differentiated antibody modality at a moment when Roche's Genentech relationship and growing biologics ambitions made platform redundancy worth the price.
