Chronic lymphocytic leukemia is the most common adult leukemia in Western countries, characterized by clonal expansion of mature CD20-positive B-cells. Median diagnosis age is 71; the disease is incurable outside of allogeneic transplant but generally indolent, with a 5-year relative survival of ~90%. CD20-directed antibodies have been a backbone of therapy since rituximab's late-1990s approval.

Competitive Landscape

At the time of the 2005 GlycArt deal, Genentech/Roche's Rituxan (rituximab) was the dominant CD20 antibody for B-cell malignancies, with Bayer/IDEC's Zevalin and Corixa/GSK's Bexxar as radio-immunoconjugate alternatives. The competitive question was who would build the next-generation CD20 follow-on: GlaxoSmithKline/Genmab's ofatumumab (Arzerra, type I, fully human) and Immunomedics' veltuzumab were already advancing. The deal effectively gave Roche a defensible Rituxan successor for CLL/iNHL and locked in glycoengineering IP applicable across antibody franchises.
