On March 25, 2024, Nuvation Bio agreed to acquire AnHeart Therapeutics in an all-stock transaction. The structure was chosen to preserve Nuvation Bio's cash balance and fund development of both the acquired and existing pipeline without a near-term capital raise. The acquisition added taletrectinib, a next-generation ROS1 inhibitor with Breakthrough Therapy designations then completing two pivotal studies in ROS1-positive non-small cell lung cancer, and safusidenib, a mutant IDH1 inhibitor in a global Phase 2 study in grade 2 and 3 IDH1-mutant glioma. Nuvation Bio described the transaction as transforming it into a late-stage global oncology company with the potential to become a commercial organisation by the end of 2025. Closing was expected in Q2 2024, subject to AnHeart shareholder approval and customary conditions. No transaction value was disclosed.

ROS1 gene rearrangements define a small, biomarker-selected subset of non-small cell lung cancer. The population skews younger and toward never-smokers, and central nervous system metastases are common, which makes brain penetration a key differentiator among ROS1 inhibitors.
Crizotinib (multi-kinase ALK/ROS1/MET inhibitor) and entrectinib (ROS1/NTRK/ALK inhibitor with CNS activity) were the established first-line options, with repotrectinib (next-generation ROS1/TRK inhibitor active against the solvent-front G2032R resistance mutation) emerging as the principal competitor. Taletrectinib competes on the same axes: CNS penetration and activity against G2032R.
IDH1-mutant gliomas are a molecularly defined subset of diffuse glioma occurring predominantly in younger adults, with a more indolent natural history than IDH-wildtype glioblastoma but near-universal eventual progression. Mutant IDH1 produces the oncometabolite 2-hydroxyglutarate, which drives epigenetic dysregulation.
Vorasidenib (dual mutant IDH1/2 inhibitor) established brain-penetrant IDH inhibition as a treatment paradigm in grade 2 glioma, alongside ivosidenib (selective mutant IDH1 inhibitor). Safusidenib competes as a selective mutant IDH1 inhibitor in the same brain-penetrant class.
