The deal targets HER2-expressing and broader solid tumor indications with a novel myeloid-activating ISAC modality. Solid tumors collectively comprise approximately 90% of US cancer incidence, with the American Cancer Society projecting roughly 1.9 million new cancer diagnoses and 608,570 deaths in 2021. HER2-positive cancers — the lead immunology focus for the BDC-1001 program — span breast (HER2+ ~15-20% of ~280,000 annual US breast cancers per ACS), gastric/GEJ (HER2+ ~15-20% of ~26,000 annual cases), endometrial, biliary tract, colorectal and salivary gland subsets. While HER2-targeted ADCs Enhertu (trastuzumab deruxtecan, Daiichi Sankyo/AstraZeneca, FDA 2019) and Kadcyla (T-DM1, Genentech, FDA 2013) and HER2 antibodies Herceptin and Perjeta have transformed HER2+ care, resistance, brain metastases, and HER2-low/heterogeneous expression remain unmet needs. The broader rationale for ISAC technology is that traditional ADCs deliver a cytotoxic payload to tumor cells but do not durably engage anti-tumor innate immunity, while checkpoint inhibitors require pre-existing immune infiltrate. ISACs combine antibody-mediated tumor targeting with TLR7/8-driven myeloid activation in the tumor microenvironment, aiming to convert immunologically cold tumors into hot ones and produce durable T-cell-dependent immunity beyond payload-mediated cell killing.

Competitive Landscape

At deal date (June 2021) the ISAC space was nascent and largely preclinical/early Phase 1.
