Zinc-finger nucleases (ZFNs) are engineered DNA-binding proteins fused to a FokI cleavage domain that introduce site-specific double-strand breaks, enabling targeted gene knockout, correction or insertion. ZFN-based therapeutics target genetic and acquired diseases — including HIV (CCR5 disruption), sickle cell, beta-thalassemia and lysosomal storage disorders — by editing patient cells ex vivo or in vivo.
At the time of the 2001 Gendaq acquisition, Sangamo's ZFP/ZFN platform competed primarily against meganucleases (Cellectis) and conventional gene therapy approaches (vector-mediated gene addition by Avigen, Targeted Genetics, GenVec). The Gendaq deal — a $30-40M stock acquisition of the UK Medical Research Council spin-off co-founded by Nobel Laureate Aaron Klug — folded in 22 patent applications, two issued patents, 16 scientists and roughly $6M cash, and is widely credited with letting Sangamo 'corner the market on the use of ZFPs in gene editing'. What the deal changed: it eliminated Sangamo's only direct ZFP IP rival and turned the platform into a licensable franchise, ultimately enabling partnerships with Pfizer (sickle cell, hemophilia A), Sanofi (beta-thalassemia), Biogen and Shire.
