Alzheimer's disease is a progressive neurodegenerative disorder and the most common cause of dementia. Recently approved amyloid-targeting antibodies slow decline modestly but carry safety and access burdens, leaving substantial unmet need for symptomatic and disease-modifying small molecules that improve cognition.
Approved anti-amyloid antibodies lecanemab (Eisai/Biogen) and donanemab (Eli Lilly) clear amyloid plaques; cholinesterase inhibitors and memantine remain symptomatic standards. ACD856 pursues a differentiated neurotrophin (Trk) positive-allosteric-modulation mechanism.
