Companion diagnostics identify which tumors carry mutations that predict response or resistance to targeted therapy. KRAS mutations (~40% of metastatic colorectal cancers) confer resistance to anti-EGFR antibodies (cetuximab, panitumumab); EGFR-activating mutations in NSCLC predict response to TKIs (erlotinib, gefitinib).
