Key facts

Disease & market context

Solid Tumors (DARPin Platform)

1.9M US cases/yr · $200.0B Global Oncology (2024)

Disease Overview

Novartis licensed MP0310, a bispecific DARPin targeting FAP and 4-1BB for tumor-localized immune activation.

Platform Landscape

DARPins compete with antibody bispecifics. FAP-targeted approach aims for tumor-selective activation.

COVID-19 Therapeutics (DARPin)

Competitive Landscape

Ensovibep (MP0420) is a multi-specific DARPin binding three epitopes on SARS-CoV-2 spike RBD; the COVID-19 therapeutics landscape at development is MOA-segmented. Anti-spike monoclonal antibodies: Regeneron's REGEN-COV (casirivimab/imdevimab), Lilly's bamlanivimab/etesevimab, GSK/Vir's sotrovimab (Xevudy), and Lilly's bebtelovimab all received EUAs but had EUAs successively revoked or limited as Omicron subvariants (BA.2, BA.4/5, XBB) eroded neutralization. AstraZeneca's Evusheld (tixagevimab/cilgavimab) was de-authorized in January 2023. Oral small-molecule antivirals: Pfizer's Paxlovid (nirmatrelvir/ritonavir) dominates outpatient treatment with ~$12.5B 2022 revenue (peaking) declining to ~$1.2B in 2024; Merck's Lagevrio (molnupiravir) remains secondary. IV antivirals: Gilead's Veklury (remdesivir) holds the hospitalized niche (~$3.9B 2022 revenue). Commercial threat sizing for ensovibep: DARPin's multi-epitope design theoretically preserves activity against variants that escape mAbs, but the ACTIV-3 trial readout in 2022 was mixed, Novartis returned rights in 2022, and the category has collapsed as Paxlovid captured outpatient share and all variant-vulnerable mAbs were withdrawn. Residual commercial opportunity is minimal absent a pandemic-scale new variant.
