Disease Overview

Autoimmune diseases driven by autoreactive B cells — including lupus, systemic sclerosis and myositis — have shown deep, drug-free remissions after autologous CD19 CAR-T "immune reset." Ex vivo CAR-T requires leukapheresis, lymphodepleting chemotherapy and complex manufacturing, which limits use in non-oncology populations. In vivo CAR-T aims to deliver the CAR construct directly to circulating T cells, removing cell manufacturing entirely.

Competitive Landscape

In vivo CAR-T is a crowded emerging field: Capstan Therapeutics (acquired by AbbVie) uses targeted lipid nanoparticle-delivered mRNA to CD8+ T cells; Umoja Biopharma uses lentiviral VivoVec particles; Kelonia and Interius are also pursuing viral in vivo delivery.
